I suffer from bipolar depression that has been all but refractory to treatment. I did respond to memantine (Namenda), but only for a short while. That I responded to it at all is probably an indication that it will work well for at least some people. Memantine has been used in Germany for over a decade, and I have not seen any reports of harmful long term adverse effects. It is also used for Parkinsons there. I think it is a very safe drug that usually produces few side effects. I don't know if memantine will be found to be most beneficial as monotherapy or as an augmentor of other drugs. The target dosage for Alzheimers is 20mg. However, dosages of 40mg are currently being used to treat neuropathies.
