I'm always on the lookout for any information on adrafinil and/or modafinil & how they work. I came across this article on adrafinil from the CNS Drug Reviews. It was written by Norton W. Milgram, Heather Callahan, & Christina Siwak of the Division of Life Sciences, University of Toronto at Scarborough, Toronto, Ontario, Canada. It's very well written & is not difficult to understand. If it seems too long for anyone, you could just go to the Conclusions & Summary at the end & read that but you'll miss some good information.
I found it very interesting how it hypothesised that adrafinil may have three possible modes of action - as a selective a1-adrenergic receptor agonist, by modifying the intracerebral release of amino acids (both GABA & glutamate), & by increasing cerebral metabolism in certain areas of the brain. Adrafinil is also metabolised into two different metabolites, one an amide & one an acid. One of the metabolites of adrafinil is modafinil itself!
It also discussed 6 separate studies done on human volunteers in hospitals in France & the correlation betweeen similar effects among all 6 studies. I honestly think that both adrafinil & modafinil will prove to be very promising drugs due to their widespread effect on different neurotransmitters & their mode of action. I find it to be quite the wonder drug for me but people need to be aware that it doesn't have immediate effects for most, & if it does, it may be placebo-induced. Maximum effects occured around day 45 of treatment but continued to increase even afterwards.
