New antidepressants in the pipeline?

Posted by SLS on March 11, 2023, at 10:59:44

There are novel antidepressants coming up the pipieline - finally.

Here are two that should be out soon. Each uses a different mechanism of action.

------------------------------------------------

ESMETHADONE - NMDA receptor antagonist

Relmada's lead program, REL-1017 (esmethadone), is a new chemical entity (NCE) and novel NMDA receptor (NMDAR) channel blocker that preferentially targets hyperactive channels while maintaining physiological glutamatergic neurotransmission.

------------------------------------------------

ZURANOLONE - positive allosteric modulator of GABA receptors.

Zuranolone is an investigational drug being evaluated as a 14-day, rapid-acting, once-daily, oral treatment in adults with MDD and PPD. The application has been granted priority review and the FDA has assigned a Prescription Drug User Fee Act (PDUFA) action date of August 5, 2023.Feb 6

There are several others that look good. Now, no one can say that they have tried every drug.

Please wait for these novel compounds to work their way up the ladder. It is not certain that you will fail to respond to drugs you have never tried before, and whose mechanisms of action are novel to your brain. There is a new potent dopamine reuptake inhibitor coming out with relatively low potencies at the NE and 5-HT transporters. At the moment, I can't remember its name.

We were in a slump for 20 years. I like having options, too, despite my currently being in remission. Sometimes antidepressants stop working for no obvious reason.
