Posted by ace on December 25, 2009, at 1:12:34

Looked into this a little, I can't see much evidnece of low dose Amisulpride (12.5-100mg) causing much in terms of weight gain...

Can anybody relay their experience with Amisulpride at this dose with regards to weight increase?

Cheers!

Hi Ace,

I've never been on amisulpride longer than several weeks so I cannot attest to whether it increases weight or not. I am a hard gainer anyway and didn't even gain much from zyprexa.

But I can say it increased my appetite, even at doses of 12.5mg per day, 25mg per day, or 25mg every other day. The lack of appetite I had from either Savella or Nortriptyline was mostly reversed by the addition of a tiny bit of amisulpride. For the first time in a long time I actually got the munchies. I think lately it has been theorized that in addition to its unique d2/d3 mechanism it may also be a 5ht7 antagonist, and who knows what other minor mechanisms it might have.

Experts claim it is the antihistamine action of drugs that increases appetite. Though I'm sure it does in many people, I disagree with that as being an accurate general statement. D2/D3 antagonism, either pre or post, and antagonism of 5ht receptors I think are big players as well as antihistamine.

Remeron and Nortriptyline are both potent antihistamines and actually lessened my appetite a lot, so obviously mileage varies.

From surfing the web a lot over the years on amisulpride, I cannot recall hardly any mention of anyone gaining weight on it. I would suspect it is probably the most weight-friendly one out there. Maybe at high doses it might have more risk. At low doses it is more of a "mild relaxed stimulation". For me anyway. I take it now and then during real bad times, but not regularly. It begins to feel more like any other antipsychotic if I take it continually. I don't think that happens with anyone else, I am just very sensitive.

To sum it up, I would expect weight gain risk on low dose amisulpride to be very low.
