I saw the following statement and wondered if there was a way around this:
"My theory is that, because it's an agonist, the brain down-regulates the dopamine receptors to compensate, so eventually the benefit wears off."

The downregulation does not mean it wears off. Infact the alteration in receptors can occur when the drug works.
For instance. SSRI's downregulate the 5-ht1a receptors, but this is one of the things that is believed to be associated with the responce. Ie post mortem brains have shown to have significantly upregulated 5-ht1a receptors.

Linkadge,
Hope you are feeling better. How do they know that SSRIs down regulate 5-ht1a can they actually see that happening with scans? Also why would reducing the number of 5-ht1a receptors have an affect on depression?
I always seem to be on here to ask questions but it's only because there are so many people on here who seem so much more knowledgeable on this subject. I never even did chemistry at school and so feel way out of my depth sometimes.
