"TC-5214 -- viewed by analysts as a high-risk, high-reward project -- is designed to work in a novel way by modulating neuronal nicotinic receptors in the brain. Over-stimulation of these receptors is thought to be associated with depression."
The key word here is "associated". This over-stimulation could be a consequence rather than a cause of depression. This may not have been ascertained prior to designing the drug, making its development very risky financially. Let's hope that this failure doesn't make the drug company shy to test novel drugs in the future.
