The think is, desipramine (a major metabolite of imipramine) is pharmacologically quite different to imipramine. Desipramine is a selective NRI with low affinity for other receptors eg. alpha receptors, muscarinic receptors etc. Imipramine has a high affinity for both alpha receptors and muscarinic receptors.

In contrast to this, both venlafaxine and desvenlafaxine are 'simply' serotonin and NE reuptake inhibitors with (presumably) very similar pharmacological effects.
